Cultures of lymph node cells from the animal that received thymectomy before irradiation (XRT) showed an increase in [3H]-thymidine incorporation, when compared with animals that received thymectomy and irradiation (TXIR), or the placebo vaccine (V). An increase in migration was seen with XRT cultured cells as compared with V and TXIR cultured cells. Migration of V and TXIR lymph node cells was significantly less than untreated control cells. Cells from XRT animals added thymus extract or Con A to migration assays enhanced migration of XRT cells. Cells from TXIR animals added Con A did not enhance the migration of TXIR cells. XRT cells added Con A produced a decrease in migration that was not seen with TXIR cells. XRT cells had increased migration to zymosan, whereas Con A induced migration was decreased. Con A reactivity of XRT lymph node cells was slightly increased when compared with V and TXIR cells, but the XRT Con A reactivity was significantly decreased when compared with V and TXIR cells. The Con A reactivity of V and TXIR cells was increased, but this was not a significant difference. The same antigenic molecules present on Con A-reactive cells of V and TXIR animals were detected on Con A reactivity of XRT cells, and the density of binding on these molecules was decreased. These results suggest that in the thymectomized irradiated animal, the thymic environment resulting from irradiation and the decreased level of thymic function are factors that induce a change in the immune response of the animal.Q:
